چکیده

The human promyelocytic leukemia cell line. HL-60. undergoes differentiation in the presence of dimethyl sulfoxide (DMSO). phorbol esters, and other agents. Studies were undertaken to determine whether agents that promote differentiation influence membrane fluidity and to assess whether changes in membrane fluidity occur during the process of differentiation. Cells were grown in the presence of DMSO, 1 .25%. or tetradecanolyl-phorbol 1 3-acetate (TPA). 1 .6 x 10 5M. Surface membranes were isolated from cells by sucrose density sedimentation following cell disruption with nitrogen cavitation. Membrane fluidity was assessed by the fluorescence polarization of the membrane probe 1.6diphenyl-1 .3,5-hexatriene (DPH). A progressive decrease in membrane fluidity was first observed on day 3 following exposure of HL-60 cells to DMSO. and this reached a maximum on day 5. coincident with morphological and cytochemical differentiation. Similar changes in membrane fluidity were observed in liposomes prepared from extracted membrane lipids. The ratio of membrane cholesterol to phospholipid was increased 37% on day 5, and there was a small increase in the ratio of saturated to unsaturated fatty acids, with no significant change in phospholipid composition. No changes in membrane fluidity were observed during 1 8 hr of exposure of HL-60 cells to TPA, after which these cells became glass adherent. These studies demonstrate that significant changes in membrane lipid composition and fluidity occur during the process of differentiation in vitro. However. agents that stimulate leukemic cells to differentiate do not themselves appear to perturb bulk membrane fluidity.

Assessment of Membrane Fluidity Fluctuations during Cellular Development Reveals Time and Cell Type Specificity

Cell membrane is made up of a complex structure of lipids and proteins that diffuse laterally giving rise to what we call membrane fluidity. During cellular development, such as differentiation cell membranes undergo dramatic fluidity changes induced by proteins such as ARC and Cofilin among others. In this study we used the generalized polarization (GP) property of fluorescent probe Laurdan us...

The lipid composition and membrane fluidity of Dictyostelium discoideum plasma membranes at various stages during differentiation.

There are only minor changes in the amounts of the major lipid constituents of Dictyostelium discoideum plasma membranes during the early stages in the differentiation of this organism. By the time cells reach the pseudoplasmodial stage of development there are small increases in the amounts of phosphatidylinositol, phosphatidylglycerol, and lysophosphatidylethanolamine, and small decreases in ...

Effects of alcohol on membrane fluidity of human erythrocyte.

Membrane fluidity in human erythrocytes was measured by a spin label method using an electron spin resonance spectrometer in healthy volunteers after ingestion of alcohol (1.5 ml of whisky/kg body weight). Fluidity in the lipid bilayer closer to the hydrophilic face decreased at 30 min and 90 min, and fluidity in the hydrophobic core decreased at 90 min after ingestion of alcohol. In the same e...

Assessment of Membrane Fluidity in Individual Yeast Cells by Laurdan Generalised Polarisation and Multi-Photon Scanning Fluorescence Microscopy

Here we describe techniques that we developed for monitoring membrane fluidity of individual yeast cells during environmental adaptation and physiological changes. Multiphoton scanning fluorescence microscopy using laurdan as a membrane probe enables determination whether fluidity changes seen by spectroscopy reflect universal responses or changes only of sub-populations. Yeast membranes are a ...
